Ribosome-inactivating proteins (RIPs) are enzymes that inhibit protein synthesis after depurination of a specific adenine in rRNA. The RIP family members are classified as type I RIPs that contain an RNA-N-glycosidase domain and type II RIPs that contain a lectin domain (B chain) in addition to the glycosidase domain (A chain). In this work, we identified 30 new plant RIPs and characterized 18 Ricinus communis RIPs. Phylogenetic and functional divergence analyses indicated that the emergence of type I and II RIPs probably occurred before the monocot/eudicot split. ABSTRACT The effect of substituting soybean meal (SM) with four levels (0, 33, 67 and 100%) of detoxified castor cake (DCC) on intake, nutrient digestibility and nitrogen balance was evaluated in uncastrated, male, crossbred sheep in a randomised complete block design with four treatments and five replications. There was a decreasing linear effect from the levels of substitution on dry matter (DM), organic matter (OM) and crude protein (CP) intake, total carbohydrates (TC), non-fibrous carbohydrates (NFC) and total digestible nutrients (TDN), and on DM, OM and TC digestibility, for reductions of 2.04, 1.70, 0.35, 1.56, 1.89 and 1.67 g dia-1 and 0.34, 0.53 and 0.51 g kg-1 DM respectively, for each 1% SM substituted with DCC. The effect of replacing of 0; 0.33; 0.67 and 1.0 (kg/kg) of soybean meal (SBM) by undecorticated castor seed meal treated with calcium oxide (CMT - 60 g/kg) was evaluated on performance and efficiency of nutrient utilization in dairy cows. Sixteen Holstein and crossbred cows were distributed in four 4 × 4 latin squares. Animals received concentrated feed at a ratio of 1 kg for 3 kg of milk produced, in the natural matter. The diets had the same amount of nitrogen (150.4 g crude protein/kg DM), containing 325.6 g of concentrated feed/kg DM. Ricinus communis L. is of great economic importance due to the oil extracted from its seeds. Castor oil has been used for pharmaceutical and industrial applications, as a lubricant or coating agent, as a component of plastic products, as a fungicide or in the synthesis of biodiesel fuels. After oil extraction, a castor cake with a large amount of protein is obtained. However, this by-product cannot be used as animal feed due to the presence of toxic (ricin) and allergenic (2S albumin) proteins. Here, we propose two processes for detoxification and allergen inactivation of the castor cake. Lethal Toxin Neutralizing Factor (LTNF), isolated from opossum with having molecular weight 63 kDa, is a potent antidote for animal, plant, and bacterial toxins. This communication deals with the identification of a small fragment of LTNF eliciting the anti-lethal activity of animal, plant, and bacterial toxins when tested in mice. Purified LTNF was treated with trypsin to cause fragmentation at the arginine and lysine sites. The fragments were separated by high-performance liquid chromatography (HPLC) and were tested against anti-LTNF for binding affinity by enzyme-linked immunosorbent test (ELISA).
